ABSTRACT

Abstract Background: Adaptive immune cells, including CD4+CD69+ and CD4+CD25+FoxP3+ regulatory T (Treg) cells, are important for maintaining immunological tolerance. In human systemic lupus erythematosus (SLE), CD4+CD25+FoxP3+ Treg cells are reduced, whereas CD69 expression is increased, resulting in a homeostatic immune imbalance that may intensify autoreactive T cell activity. To analyze the mechanisms implicated in autotolerance failure, we evaluated CD4+CD69+ and CD4+CD25+FoxP3+ T cells and interleukin profiles in a pristane-induced SLE experimental model. Methods: For lupus induction, 26 female Balb/c mice received a single intraperitoneal 0.5 ml dose of pristane, and 16 mice received the same dose of saline. Blood and spleen samples were collected from euthanized mice 90 and 120 days after pristane or saline inoculation. Mononuclear cells from peripheral blood (PBMC), peritoneal lavage (PL) and splenocytes were obtained by erythrocyte lysis and cryopreserved for further evaluation by flow cytometry using the GuavaEasyCyte TM HT. After thawing, cells were washed and stained with monoclonal antibodies against CD3, CD4, CD8, CD25, CD28, CD69, FoxP3, CD14 and Ly6C (BD Pharmingen TM). Interleukins were quantified using Multiplex® MAP. The Mann-Whitney test and the Pearson coefficient were used for statistical analysis, and p < 0.05 considered significant. Results: Compared with the controls, SLE-induced animals presented increased numbers of CD4+CD69+ T cells in the blood on T90 and T120 (p = 0.022 and p = 0.008) and in the spleen on T120 (p = 0.049), but there were decreased numbers in the PL (p = 0.049) on T120. The percentage of Treg was lower in blood (p < 0.005 and p < 0.012) on T90 and T120, in spleen (p = 0.043) on T120 and in PL (p = 0.001) on T90. Increased numbers of CD4+ CD69+ T cells in the PL were positively associated with high IL-2 (p = 0.486) and IFN-γ (p = 0.017) levels, whereas reduced Treg cells in the blood were negatively correlated with TNFα levels (p = 0.043) and positively correlated with TGFβ1 (p = 0.038). Conclusion: Increased numbers of CD4+CD69+ T cells and reduced numbers of CD4+CD25+FoxP3+ Treg cells with an altered interleukin profile suggests loss of autotolerance in pristane-induced lupus mice, which is similar to human lupus. Therefore, this model is useful in evaluating mechanisms of cellular activation, peripheral tolerance and homeostatic immune imbalance involved in human SLE.

In Aggregatibacter actinomycetemcomitans, different serotypes have been described based on LPS antigenicity. Recently, our research group has reported a differential immunogenicity when T lymphocytes were stimulated with these different serotypes. In particular, it was demonstrated that the serotype b of A. actinomycetemcomitans has a stronger capacity to trigger Th1- and Th17-type cytokine production.Objective This study aimed to quantify the expression of different CC chemokines (CCLs) and receptors (CCRs) in T lymphocytes stimulated with the differentA. actinomycetemcomitans serotypes. In addition, the expression of the transcription factors T-bet, GATA-3, RORC2, and Foxp3, master-switch genes implied in the Th1, Th2, Th17, and T-regulatory differentiation, respectively, was analysed in order to determine T-cell phenotype-specific patterns of CCL and CCR expression upon A. actinomycetemcomitans stimulation.Material and Methods Human naïve CD4+ T lymphocytes were obtained from healthy subjects and stimulated with autologous dendritic cells primed with the differentA. actinomycetemcomitans serotypes. The expression levels for the chemokines CCL1, CCL2, CCL3, CCL5, CCL11, CCL17, CCL20, CCL21, CCL25, and CCL28, as well as the chemokine receptors CCR1, CCR2, CCR3, CCR4, CCR5, CCR6, CCR7, CCR8, CCR9, and CCR10 were quantified by qPCR. Similarly, the expression levels for the transcription factors T-bet, GATA-3, RORC2, and Foxp3 were quantified and correlated with the CCL and CCR expression levels.Results Higher expression levels of CCL2, CCL3, CCL5, CCL20, CCL21, CCL28, CCR1, CCR2, CCR5, CCR6, CCR7, and CCR9 were detected in T lymphocytes stimulated with the serotype b of A. actinomycetemcomitans compared with the other serotypes. In addition, these higher expression levels of CCLs and CCRs positively correlated with the increased levels of T-bet and RORC2 when T lymphocytes were stimulated with the serotype b.Conclusion A T-lymphocyte response biased towards a Th1- and Th17-pattern of CCL and CCR expression was detected under stimulation with the serotype b ofA. actinomycetemcomitans.

ABSTRACT

El asma es una enfermedad atópica caracterizada por un incremento de los eosinófilos y de la inmunoglobulina E (IgE) en la mucosa nasal y en la sangre periférica. Con los avances en el conocimiento de la fisiopatología y de las técnicas de laboratorio, en estos momentos se pueden determinar algunas proteínas de la superficie de los linfocitos T llamadas CD, por ejemplo CD62L, CD152, CD11a. Material y métodos: se reclutaron 15 sujetos distribuidos en tres grupos de cinco personas (control, asma leve y asma severa). Se determinaron CD11a, CD30, CD62L. Se efectuó un Análisis con métodos estadísticos no paramétricos. Resultados: se encontraron diferencias con respecto a CD62L (p = 0.009) relacion ndose con asma severa. Conclusión: se demuestra la existencia de un cambio en la expresión de los CD. Hace falta un número mayor de pacientes para apoyar aún más nuestros resultados.

INTRODUÇÃO: O aborto recorrente é um grande enigma, pois apesar de várias causas que lhe säo atribuidas poucas tem um real valor. O papel do fator imunológico tem sido pesquisado amplamente. OBJETIVO: Avaliar alguns testes imunológicos em pacientes com aborto recorrente. TIPO DE ESTUDO: prospectivo LOCAL: Setor de Reproduçäo Humana do Departamento de Ginecologia e Obstetrícia da Faculdade de Medicina de Ribeiräo Preto. PARTICIPANTES: Num estudo piloto foram selecionados nove casais com duas perdas fetais ou mais sem causa aparente e nove casais voluntários com pelo menos dois filhos, sem história de aborto e com idade inferior a 40 anos VARIAVEIS ESTUDADAS: A freqüência de compartilhamento de antígenos HLA e de anticorpos linfocitotóxicos contra antígenos paternos foram avaliados por métodos sorológicos, a variaçäo de fenótipos celulares (CD4, CD8, CD19, CD16, CD56 e HLA-DR) por citometria de fluxo, a atividade "natural killer" (NK) pela liberaçäo de 51Cr e a dosagem de progesterona por radioimunoensaio. RESULTADOS: Näo houve diferença de compartilhamento entre casais com aborto recorrente tanto em relaçäo à classe I quanto II e näo se detectou o aparecimento de anticorpos citotóxicos no grupo investigado. Houve um número absoluto maior de células CD8+(587 vs 448 linfócitos/mmü, p=0,01) e das CD19+(215 vs 182 linfócitos/mmü, p=0,05) nas pacientes. A atividade NK näo foi estatisticamente diferente entre os dois grupos estudados, mas houve uma tendência à reduçäo da atividade NK entre pacientes com aborto recorrente. Näo houve correlaçäo da atividade NK com um número de células CD16+ e CD56+ nem com dosagem de progesterona nos dois grupos estudados. CONCLUSÃO: Estes dados sugerem que o compartilhamento de antígenos HLA, o aparecimento de anticorpos linfocitotóxicos e elevaçäo da atividade NK podem näo ser importantes para a ocorrência de abortos repetidos. o aumento de células CD8+ e CD19+ circulantes pode ocorrer independente de qualquer terapêutica e a citotoxicidade contra antígenos fetais pode ser mediada por células T e näo por células NK.

ABSTRACT

This study reviewed 65 cases of polymorphic reticulosis (PR) with respect to clinical and histopathologic bases, and immunohistochemical studies were done using MT1 and UCHL as T-cell markers, MB2 as a B-cell marker and alpha-1-antichymotrypsin as a histiocytic marker. The results obtained were as follows: 1. The male to female ratio was 2.4:1 and the mean age of patients was 44.5 years. The sites involved primarily were the nasal cavity, tonsil and pharynx and about one-fourth of the total cases showed extensive involvement of two anatomical sites at initial presentation. 2. Almost all cases showed characteristic histologic features similar to those of peripheral T-cell lymphoma and showed positive reaction to the T-cell marker. The above immunohistochemical findings suggest strongly that quite a significant portion of PR is in fact T-cell lymphoma.

En un modelo experimental de inmunodeficiencia provocada por déficit de proteínas, se demuestra: 1) en el timo, existencia de graves alteraciones en sus poblaciones celulares que se encuentran drmáticamente disminuídas: W3/13+ (pan- T), W3/25+ (CD4+) y OX8+ (DC8+), existiendo una población celular que contiene TdT como único marcador; 2) alteraciones en el ciclo celular de la IgA que permitieron demostrar in vivo la presencia de células pre-B en placas de Peyer con alteración marcada en las subpoblaciones T; 3) los mecanismos involucrados en la inmunidad mediada por células son dañados por el déficit de proteínas y continúan alterados después de que dicho déficit ha sido superado; 4) en el timo, la administración por via oral de una dieta de caseína al 20% restaura la población celular- T W3/13+ luego de 21 días, pero la subpoblación W3/25+ (CD4+) se encuentra muy disminuida; 5) al mismo tiempo se reinicia, en placas de Peyer, la diferenciación de los precursores de células plasmáticas de clase IgA, aunque se encuentra la diferenciación de los precursores de células plamáticas de clase IgA, aunque se encuentra alterado, tanto en ellas como en ganglio mesentérico, el número absoluto de células que expresan y contienen IgA, así como el de las células T maduras, especialmente la población CD4+; 6) el dearrollo de un mecanismo de tolerancia a dextrina que es antígeno-específico, está mediado por células-T y puede ser transferido a receptores normales, ya sea con células provenientes de...

ABSTRACT

Using anti-T6 and anti-HLA-DR monoclonal antibodies, this study was designed to attain what would do to epidermal Langerhans cell (LC) markers in psoriatic patients when two remedies, etretinate and UVB, having controversial effects on LC were put to use simultaneously. In normal and psoriatic subjects, HLA-DR+ LC was approximately 80% of T6+ LC and a single dose of UVB equivalent to minimal erythema dose (MED), reduced LC membrane markers to approximately 30% of non-irradiated control. The recovery of LC membrane markers, after a single dose of UVB exposure were significantly faster in the group of etretinate treated psoriatic subject than only UVB irradiated psoriatic control. Taken together, seemed to exert prompt recovery of normalization of the number of LC from the depletion following the UVB.
